Tirzepatide is a single peptide that activates both GIP and GLP-1 receptors, combining two gut-hormone signals5.
Glucose-dependent insulin secretion lowered HbA1c by up to 2.30 percentage points, more than semaglutide in SURPASS-22.
It delays gastric emptying, and an albumin-binding fatty-acid side chain extends its half-life to support once-weekly dosing5.
What the research shows
SURMOUNT-1’s top dose averaged 20.9% weight loss at 72 weeks versus 3.1% with placebo, and 57% on 10–15 mg lost a fifth of their weight.
In SURMOUNT-5, tirzepatide produced 20.2% weight loss versus 13.7% with semaglutide at 72 weeks.
In SURPASS-2, all three doses lowered HbA1c more than semaglutide, with greater weight reduction.
In SURMOUNT-4, 89.5% of people who stayed on tirzepatide kept at least 80% of their weight loss at 88 weeks, versus 16.6% on placebo.
SURMOUNT-5 randomized 751 adults; mean reductions at 72 weeks were 20.2% versus 13.7% with semaglutide at the studied doses.
