In healthy men, an infusion produced one discrete GH episode peaking at 0.67 hours, with a 2-hour terminal half-life2.
In swine it released GH without significant ACTH or cortisol release, even at doses more than 200-fold above its effective GH dose1.
Ghrelin receptors also govern gastrointestinal transit. After abdominal surgery in rats, repeated ipamorelin increased bowel output, food intake and weight gain4.
What the research shows
Its discovery study separated a strong GH response from ACTH and cortisol release, setting it apart from GHRP-6 and similar compounds.
Healthy volunteers showed a single GH peak at 0.67 hours with a short 2-hour terminal half-life.
In rats, ipamorelin’s plasma clearance was 5-fold lower than GHRP-6, with about 20% intranasal bioavailability.
After surgery in rodents, repeated dosing increased bowel output, food intake and body-weight gain.
In 114 postoperative patients, median time to a first tolerated meal was 25.3 h with ipamorelin versus 32.6 h with placebo, a difference that was not statistically significant.
